DNN uses Skins for the general page layout, which include vertical panes which contain the content via modules.
for an individual home page layout, you would either create an individual skin or a skin varient, depending of your specific needs.

The homepage is the exception when you apply a theme.
You can still alter the theme by going to the page settings - appearance tab

This video on theme management in DNN should be helpful to you.  (It's a bit old, but still very relevant.)
